Safety: If any solutions get into a student9s eyes, flush with water for 15 minutes. Complete an Accident
Report.
Materials required:
Digital water bath
Ice or ice maker
95% ethanol
Permanent markers
Waste beakers
Tube racks for 15 mL tubes
Tube racks for 1.5 mL tubes
Lysis buffer (110 mL)
Powdered protease and salt 1.5 g
15 mL tubes
1.5 mL tubes
Transfer pipets
Helix vials, screw caps, and cords
Prep:
Set the digital water bath to 50°C
Place a 15 mL tube rack into the water bath
Pour dH20 into the ice maker and turn on
Place 380 mL of 95% ethanol in the freezer at least 1 hour prior to use (pour in a beaker and parafilm)
Pour protease powder into a 15 mL tube and add 15 mL of dH20. Label 8Protease 100 mg/mL9 and date
(Good for 1 week at 4°C)
Aliquot 1.25 mL of rehydrated protease into nine 1.5 mL tubes. Label the tubes 8prot9
For each student, dispense 3 mL of dH20 into a 15 mL tube
Dispense 10 mL of lysis buffer into nine 15 mL tubes. Label the tubes 8lysis9
At each station (n=9), place:
Two 15 mL tubes containing the 3 mL of dH20
One 8prot9 tube
One 8lysis9 tube
Three transfer pipettes
One permanent marker
One beaker for waste
Helix vial, screw cap, and cord
Protocol:
_____Label the 15 mL tube containing 3 mL dH20 with your initials
_____Gently chew the insides of your cheeks (not butt) for 30 seconds no blood
_____Take the water from the 15 mL tube into your mouth, and swish vigorously for 30 seconds
_____Carefully expel the liquid back into the 15 mL tube
_____Add 2 mL of lysis buffer to your tube
_____Place the cap on the tube, and gently invert the tube 5 times (no shaky shaky)
_____Add 5 drops of protease (8prot9) to your tube
_____Place the cap on your tube, and gently invert it a few times
